Multiple airy beams light-sheet fluorescence microscopy

چکیده

Light-sheet fluorescence microscopy (LSFM) is a kind of volumetric imaging methodology suited for long term living specimens at high temporal-spatial resolution. A single Airy beam (SAB) light-sheet can extend the field view benefiting from its non-diffracting nature, but cost out-of-focus background and low contrast caused by side lobes illumination. Here, we propose method to generate sort multiple beams (MAB), which are linearly superimposed with different scale factors. Compared SAB light-sheet, energy more concentrated on focal plane detection objective, improve decrease photodamage effect. Furthermore, combined complementary subtraction (CBS) strategy increase axial resolution, termed as beams-complementary method, enables resolution 1.2 μm while keeping 450 × μm. The effectiveness demonstrated fluorescent beads aspergillus conidiophores.

Three Dimensional Fluorescence Imaging Using Multiple Light-Sheet Microscopy

We developed a multiple light-sheet microscopy (MLSM) system capable of 3D fluorescence imaging. Employing spatial filter in the excitation arm of a SPIM system, we successfully generated multiple light-sheets. This improves upon the existing SPIM system and is capable of 3D volume imaging by simultaneously illuminating multiple planes in the sample. Theta detection geometry is employed for dat...

Structured light sheet fluorescence microscopy based on four beam interference.

A 3D structured light sheet microscope using a four-faceted symmetric pyramid is presented. The sample is illuminated by the resulting four beam interference field. This approach combines advantages of standing wave and structured illumination microscopy. Examples of micrographs of fluorescently labeled Chinese hamster ovary (CHO) cells as well as of the compound eyes of drosophila are shown an...

Nonlinear light-sheet fluorescence microscopy by photobleaching imprinting.

We present a nonlinear light-sheet fluorescence microscopy (LSFM) scheme based on photobleaching imprinting. By measuring photobleaching-induced fluorescence decay, our method simultaneously achieves a large imaging field of view and a thin optical section. Furthermore, the scattered-light-induced background is significantly reduced, considerably improving image contrast. Our method is expected...

Using Light Sheet Fluorescence Microscopy to Image Zebrafish Eye Development

Light sheet fluorescence microscopy (LSFM) is gaining more and more popularity as a method to image embryonic development. The main advantages of LSFM compared to confocal systems are its low phototoxicity, gentle mounting strategies, fast acquisition with high signal to noise ratio and the possibility of imaging samples from various angles (views) for long periods of time. Imaging from multipl...

Hyperspectral light sheet microscopy

To study the development and interactions of cells and tissues, multiple fluorescent markers need to be imaged efficiently in a single living organism. Instead of acquiring individual colours sequentially with filters, we created a platform based on line-scanning light sheet microscopy to record the entire spectrum for each pixel in a three-dimensional volume. We evaluated data sets with varyin...
